The present invention is related to a method for the selection of recombinant clones having integrated a gene of interest (6) and a nucleotide sequence (3) encoding a functional antidote protein (4) to a toxic molecule (5), wherein said recombinant clones are the ones which survive following their integration into a host cell (20) comprising in its genome a nucleotide sequence (21) encoding said toxic molecule (5).The present invention is also related to a nucleic acid construct, a vector comprising said nucleic acid construct, a host cell and a cloning and/or sequencing kit for performing said method.
La présente invention concerne une méthode de sélection de clones de recombinaison dans lesquels sont intégrés un gène intéressant (6) et une séquence nucléotidique (3) codant une protéine antidote fonctionnelle (4) destinée à une molécule toxique (5), lesdits clones de recombinaison étant les clones qui survivent suite à leur intégration dans une cellule hôte (20) comprenant dans son génome, une séquence nucléotidique (21) codant ladite molécule toxique (5). Cette invention concerne également une construction d'acide nucléique, un vecteur comprenant cette construction d'acide nucléique, une cellule hôte et un kit de clonage et/ou de séquençage permettant de mettre en oeuvre ledit procédé.
