The invention relates to a method for the integrated treatment of biomass from a cell culture process for producing clear cell lysate containing plasmid DNA. According to the invention, in a first step filtering occurs in a filter in the presence of a filtering agent. In a second step, the biomass contained in the filter cake which is obtained is thermally decomposed and the cell lysate is filtered in another step. The obtained cell lysate containing plasmid DNA is characterised by a clarity of OD600 of at a maximum of 0.05E/cm and which is suitable for cloning, transformation, transfection, microinjection in cells, gene therapy, DNA vaccination and/or for polymerase chain reaction (PCR).
L'invention concerne un procédé pour le traitement intégré de biomasse provenant d'un processus de culture cellulaire afin de produire du lysat cellulaire clair contenant de l'ADN plasmidique. Selon l'invention, on procède lors d'une première étape à un filtrage dans un filtre en présence d'un adjuvant de filtrage, la biomasse contenue dans le gâteau de filtre obtenu est désintégrée thermiquement lors d'une deuxième étape et le lysat cellulaire est filtré lors d'une étape suivante. Le lysat cellulaire obtenu contenant l'ADN plasmidique se caractérise par une limpidité OD¿600? d'au plus 0,05 E/cm et convient au clonage, à la transformation, à la transfection, à la micro-injection dans des cellules, à la thérapie génique, à la vaccination ADN ou à la réaction en chaîne de la polymérase (PCR).
