Described herein are methods for generating recombined nucleic acids. In one method, fragments of a sequence are provided wherein the fragments have non- extendable 3' ends. A primer is provided and the primer and fragments are reacted under conditions to extend the primer to form a recombined nucleic acid molecule. In the methods herein, the non-extendable fragments act only as templates, rather than as templates and primers.

L'invention concerne des méthodes permettant de générer des acides nucléiques recombinés. Dans une méthode, on utilise des fragments d'une séquence, ces fragments possédant des extrémités 3' non étirables. On utilise également une amorce que l'on fait réagir avec les fragments dans des conditions visant à étendre l'amorce afin de former une molécule d'acide nucléique recombinée. Selon ces méthodes, les fragments non étirables ne servent que de matrices, et non pas de matrices et d'amorces.
