The invention provides biocatalytic methods for the manufacture of pure single enantiomer compounds. The invention provides methods of screening for enzymes which are highly enantioselective or enzymes that can provide any desired stereoisomer of a compound. The invention provides the use of single enantiomer substrates in performing a growth screen of a clonal library to identify highly stereoselective enzymes. In one aspect, methods for screening and identification of enzymes, e.g., transaminases, nitrilases, aldolases, epoxide hydrolases are provided. Methods for the production and screening of gene libraries generated form nucleic acids isolated from more than one organism for enzyme, e.g., transaminase, activities are also provided.
L'invention concerne des procédés biocatalytiques de fabrication de composés d'énantiomères simples purs. L'invention concerne également des procédés de criblage d'enzymes hautement énantiosélectives ou d'enzymes pouvant fournir n'importe quel stéréoisomère désiré d'un composé. L'invention concerne encore l'utilisation de substrats énantiomères simples dans la réalisation d'un écran de croissance de banque de clones permettant d'identifier des enzymes hautement stéréosélectives. Dans un aspect, l'invention concerne des procédés de criblage et d'identification d'enzymes, par exemple des transaminases, des nitrilases, des aldolases, des époxydes hydrolases. L'invention concerne enfin des procédés de production et de criblage de banques de gènes créées à partir d'acides nucléiques isolés, issus de plus d'un organisme, pour des activités enzymatiques, de transaminase par exemple.
