A biosensor for detecting a target nucleic acid, which has at least three layers, two of which are a waveguide, wherein one layer includes a nucleic acid or nucleic acid analog capable of hybridizing to the target nucleic acid, and wherein a flurophore is tethered to the nucleic acid or nucleic acid analog and wherein the biosensor functions according to direct excitation.
