Described herein is a multi-functional polymeric material for use in inhibiting adhesion and immune recognition between cells and cells, cells and tissues, and tissues and tissues. One component of the polymeric material adsorbs well to cells or tissue, and the other component of the polymeric material does not adsorb well to tissues. A water-soluble polymer that does not bear charge (polynonion) and which is not poly(ethylene glycol) is used as the non-binding component. A water-soluble polymer that is positively charged at physiological pH (polycation) and which is not poly(L-lysine) is used as the tissue binding component. When the bi-functional polymeric material contacts a tissue, the tissue-binding component binds and thus immobilizes the attached non-binding component, which will then extend generally away from the tissue surface and sterically block the attachment of other tissues. The method and compositions are useful in inhibiting formation of post-surgical adhesions, protecting damaged blood vessels from thrombosis and restenosis, and decreasing the extent of metastasis of attachment-dependent tumor cells.

L'inventon concerne un matériau polymère multifonctionnel s'utilisant pour inhiber l'adhérence et la reconnaissance commune entre des cellules, entre des cellules et des tissus et entre des tissus. Un constituant du matériau polymère présente une caractéristique d'adsorption optimisée par rapport aux cellules ou au tissu et l'autre constituant du matériau polymère ne présente qu'une caractéristique d'adsorption faible par rapport aux tissus. Un polymère soluble dans l'eau non porteur de charge (polynonion) s'utilise en tant que constituant non adhérant et un polymère soluble dans l'eau chargé positivement au pH physiologique (polycation) s'utilise en tant que constituant adhérant au tissu. Quand le matériau polymère bifonctionnel est en contact avec un tissu, le constituant adhérant au tissu adhère au constituant non adhérant fixé et, de ce fait, l'immobilise, ce dernier s'éloignant généralement de la surface du tissu et bloquant stériquement la fixation d'autres tissus. Le procédé et les compositions sont efficaces pour inhiber l'apparition d'adhérences post-chirurgicales, pour protéger des vaisseaux sanguins lésés contre la thrombose et la resténose et pour limiter l'étendue de la métastase de cellules cancéreuses dépendantes de la fixation.
