A process for the purification of an oligonucleotide synthon is provided. The process comprises subjecting an organic solution comprising an oligonucleotide synthon and lower molecular weight impurities to nanofiltration whereby the ratio of an oligonuclectide synthon to lower molecular weight impurities in the solution is increased after the nanofiltration. Preferably, the oligonucleotide synthon is a nucleoside phosphoramidite or nucleoside H- phosphonate. The nanofiltration membrane is preferably a polyimide membrane having a molecular weight cut off of 400.
L'invention concerne un procédé de purification d'un synthon oligonucléotidique. Ce procédé consiste à soumettre une solution organique comprenant un synthon oligonucléotidique et des impuretés de faible poids moléculaire à une nanofiltration, la teneur du synthon oligonucléotidique par rapport aux impuretés de faible poids moléculaire de la solution étant accrue après la nanofiltration. De préférence, le synthon oligonucléotidique est un phosphoramidite nucléosidique ou un H-phosphonate nucléosidique. La membrane de nanofiltration est de préférence une membrane polyimide présentant un seuil de coupure moléculaire de 400.
