It is found out that age-related diseases or symptoms are induced in a knockout mouse lacking the function of S1-5 gene. It is also clarified by histological analysis that the bone mineral content, the bone density and the bone strength are lowered in this knockout mouse while the osteoclast count is increased in its bone tissue. By analyzing the osteoclastogenesis ability in vitro using bone marrow-origin cells of the knockout mouse, it is also clarified that this mouse has accelerated osteoclastogenesis ability and large- sized osteoclasts compared with a wild type mouse. It is disclosed that the addition of purified S1-5 protein to this in vitro system lowers the osteoclastogenesis ability.
