The present invention relates generally to a method of modulating cytokine- mediated cellular activity and to agents useful for same. More particularly, the present invention contemplates a method of modulating tumor necrosis factor-mediated cellular activity by modulating an intracellular sphingosine kinase-dependent signalling mechanism. The method of the present invention is useful,inter alia, in the treatment and/or prophylaxis of conditions characterised by aberrant, unwanted or otherwise inappropriate cytokine- mediated cellular activity The present invention is further directed to methods for identifying and/or designing agents capable of modulating the subject sphingosine kinase-dependent signalling mechanism.
La présente invention concerne, d'une manière générale, une méthode de modulation de l'activité cellulaire induite par cytokine ainsi que des agents utiles pour ce faire. Plus particulièrement, la présente invention concerne une méthode de modulation de l'activité cellulaire induite par le facteur de nécrose tumorale par modulation du mécanisme de signalisation dépendant de la sphingosine kinase intracellulaire. La méthode de la présente invention est utile, <i>inter alia</i>, dans le traitement et/ou la prophylaxie d'états caractérisés par une activité cellulaire induite par la cytokine aberrante, indésirable ou encore inappropriée. La présente invention concerne aussi des méthodes d'identification et/ou de conception d'agents capables de moduler le mécanisme de signalisation dépendant de la sphingosine kinase en question.
