Surfactant protein D (SP-D) is a 43-kDa member of the collectin family of collagenous lectin domain-containing proteins that is er:pressed in epithelial cells of the lung. The SP-D gene was targeted by homologous recombination in embryonic stem cells that were used to produce SP-D (-¦-) mice. The SP-D (-¦-) deficiency caused inflammation, increased oxidant production by isolated alveolar macrophages, abnormal surfactant structure and levels, and decreased SP-A expression. Therefore, disclosed is the SP-D (-¦-) mouse as an excellent model for emphysema. Also included are models for testing emphysema therapies in the mouse model, methods for using SP-D protein or DNA as a treatment for emphysema and pulmonary infections, and diagnosis.

L'invention porte sur la protéine tensioactive D (SP-D), membre de 43 kDa de la famille des collectines, protéines de collagène contenant le domaine des lectines, exprimée dans les cellules épithéliales des poumons. Le gène SP-D a été ciblé par recombinaison homologue dans des cellules souches d'embryons servant à produire des souris SP-D (-/-). La déficience en SP-D (-/-) génère de l'inflammation, accroît la production d'oxydants par des macrophages alvéolaires isolés, crée des structures et niveaux tensio-actifs anormaux, et abaisse l'expression du SP-A. De ce fait, la souris SP-D (-/-) de l'invention constitue un excellent modèle pour l'emphysème. L'invention porte en outre sur des modèles permettant de vérifier les thérapies anti-emphysème dans le modèle de souris, sur des méthodes recourant à la protéine ou à l'ADN SP-D pour traiter et diagnostiquer l'emphysème et les infections pulmonaires.
