The present invention relates to methods for detecting the presence or amount of a target polynucleotide. A polynucleotide, target nucleic acid analog, and dye are combined to form a mixture. The optical property of the dye is compared to a reference value characteristics of the rate of change in the optical property of the dye in a similar mixture containing a known amount of a target polynucleotide/nucleic acid analog hybrid to determine a relative rate of change in the optical property. The relative rate of change in the optical property of dye in the mixture is correlated with the presence or amount of the specified target polynucleotide in the sample.
La présente invention concerne des procédés permettant de détecter la présence ou la concentration d'un polynucléotide cible. Dans ces procédés, un polynucléotide, un analogue d'acide nucléique cible et un colorant sont combinés pour former un mélange. Ces procédés consistent ensuite à comparer la propriété optique du colorant à une valeur de référence caractéristique du taux de variation de la propriété optique du colorant dans un mélange similaire contenant une concentration connue d'un hybride polynucléotide/analogue d'acide nucléique cible pour déterminer un taux de variation relatif de la propriété optique. Ce taux de variation relatif de la propriété optique du colorant dans le mélange est associé à la présence ou à la concentration du polynucléotide cible spécifié dans l'échantillon.
