A purified thermostable enzyme is derived from the thermophilic archaebacterium Thermococcus gorgonarius. The enzyme can be native or recombinant, retains approximately 90 % of its activity after incubation for two hours at 95 °C in the presence of stabilizing agents and possesses 3'-5' proofreading exonuclease activity. Thermostable DNA polymerases are useful in many recombinant DNA techniques, especially nucleic acid amplification by the polymerase chain reaction (PCR).
Une enzyme thermostable purifiée est dérivée de l'archaeobactérie thermophile Thermococcus gorgonarius. Il peut s'agir d'une enzyme native ou de recombinaison, qui retient environ 90 % de son activité après incubation pendant deux heures à 95 DEG C en présence d'agents de stabilisation et possède une activité d'exonucléase de correction d'épreuve en 3'-5'. Les polymérases d'ADN thermostables sont utiles dans bon nombre de techniques d'ADN de recombinaison, en particulier, dans le cas de l'amplification de l'acide nucléique par la réaction en chaîne de la polymérase.
