The present invention relates to the design of trimeric polypeptides using polypeptide structural elements derived from the tetranectin protein family, and their use in rational de novo design and production of multi-functional molecules including the application of the multi-functional molecules in protein library technology, such as phage display technology, diagnostic and therapeutic systems, such as human gene therapy and imaging. The trimeric polypeptides being constructed as a monomer polypeptide construct comprising at least one tetranectin trimerising structural element (TTSE) which is covalently linked to at least one heterologous moiety, said TTSE being capable of forming a stable complex with two other TTSEs; or as an oligomer which is comprised of two monomer polypeptide constructs as mentioned above, and which comproses three TTSEs or a multiplum of three TTSEs, or which is comprised of three monomer polypeptide constructs.

L'invention concerne l'élaboration de polypeptides trimères à partir d'éléments structurels polypeptidiques dérivés de la famille des protéines tétranectines et leur utilisation pour l'élaboration rationnelle de novo et pour la production de molécules multifonctionnelles notamment aux fins de l'application de molécules multifonctionnelles dans les technologies de banques de protéines, telles que la technologie de visualisation de phages, les systèmes de diagnostic et de traitement, notamment la thérapie génique humaine et l'imagerie. Les polypeptides trimères sont construits en tant que structure polypeptidique monomère comprenant au moins un élément structurel formant des trimères de tétranectine (TTSE), lié par covalence à au moins une fraction hétérologue, ledit TTSE étant capable de former un complexe stable avec deux autres TTSE; ou en tant qu'oligomère constitué de deux structures polypeptidiques monomères semblables à celles mentionnées ci-dessus, et comprenant trois TTSE ou un multiple de trois TTSE, ou constitué de trois structures polypeptidiques monomères.
