A method of quantitating proteins in complex samples using capillary electrophoresis can be used both to determine the concentration of a protein in a sample and to determine total protein concentration in the sample. In general, the method of determining the concentration of a marker protein comprises: (1) adding a known quantity of an internal standard compound to a sample containing at toast one protein, the internal standard compound selected from the group consisting of benzoic acid substituted with at least one halogen, producing a detector signal in relation to its concentration, and being capable of electrophoretic separation from the protein; (2) subjecting the sample and the internal standard compound to capillary electrophoresis to separate the protein and the internal standard compound from oath other and from other components in the sample; (3) measuring the detector signal produced by the internal standard compound and a detector signal produced by the protein to determine a ratio of protein signal to internal standard signal; and (4) determining the concentration of the protein in the sample from a standard curve of protein concentration versus the ratio of protein signal to internal standard signal. A typical protein measurable by the method is albumin. Typically, measurements are made at 214 em and the internal standard compound is 2,4-dichlorobenzoic acid
Un procédé de quantification des protéines dans des échantillons au moyen de l'électrophorèse capillaire peut être utilisé à la fois pour déterminer la concentration d'une protéine dans un échantillon ainsi que la concentration protéique totale. En général, le procédé de détermination de la concentration d'une protéine de référence consiste à: (1) ajouter une quantité connue de composé standard interne dans un échantillon contenant au moins une protéine, ledit composé interne standard étant choisi dans le groupe comprenant de l'acide benzoïque substitué avec au moins un halogène produisant un signal détecteur proportionnel à sa concentration, et séparable de la protéine par électrophorèse; (2) soumettre l'échantillon et le composé standard interne à une électrophorèse capillaire de manière à séparer la protéine et le composé standard interne l'un de l'autre et d'autres composants contenus dans l'échantillon; (3) mesurer le signal détecteur produit par la protéine de manière à déterminer le rapport entre le signal protéique et le signal standard interne; et (4) déterminer la concentration de ladite protéine dans l'échantillon à partir d'une courbe standard de concentration protéique selon le rapport entre le signal protéique et le signal standard. L'albumine est une des protéines typiques pouvant être mesurée avec ce procédé. Généralement, les mesures sont effectuées à 214 mn et le composé standard interne est de l'acide 2,4-dichlorobenzoïque.
