The present invention relates to a cell which is suitable for screening a candidate agent as being an inhibitor of the metabolism of tryptophan to NAD+ and/or a modulator of NAD+ levels, which cell comprises functional genes of a pathway enabling the metabolism of tryptophan to NAD+ and wherein the cell includes a copy of an exogenous gene of said pathway, from the same or different species as the cell, which exogenous gene is under the control of an inducible or constitutive promoter and wherein any endogenous copy of the gene having the same function as the exogenous gene is a non functioning gene. The present invention also relates to populations of such cells and to methods of screening candidate agents with such cells.
L'invention concerne une cellule qui permet de cribler un agent candidat inhibiteur du métabolisme de la conversion du tryptophan en NAD+ et/ou modulateur des niveaux de NAD+, laquelle cellule comprend des gènes fonctionnels d'une voie permettant le métabolisme de la conversion du tryptophan en NAD+ et comprend une copie d'un gène exogène de ladite voie, issu d'une espèce semblable à celle de la cellule ou différente de celle-ci. La cellule exogène est sous le contrôle d'un promoteur inductible ou constitutif, et toute copie endogène du gène ayant la même fonction que le gène exogène est un gène non fonctionnel. L'invention se rapporte également à des populations de cellules et à des procédés permettant de cribler les agents candidats à l'aide desdites cellules.
