ABSTRACT OF THE DISCLOSURE A plasmid which carriers an inserted regulatable promoter at a position and in an orientation-permitting transcription from the promoter into the replication region. The replication region comprises a gene coding for a gene product which is positively needed for replication. Increased transcription from the regulatable promoter leads to an increased level of the gene product which is positively needed for replication. This increased level results in a plasmid replication rate which is higher than the host cell growth rate leading to a substantial increased or uncontrolled plasmid copy number. The subject plasmids may be used as cloning vectors for the insertion of foreign (e.g. eucaryotic) DNA coding for a desired polypeptide.
