A method for improving the healing response of epithelialized tissue (e.g., skin, mucosae) to acute traumatic injury is disclosed. The method includes employing a tetracycline compound, having substantially no antibacterial activity, to improve the capacity of the epithelialized tissue to heal acute wounds. Specifically, the method involves increasing the rate of collagen accumulation of the healing epithelialized tissue above that associated with wound healing in the individual. The method decreases proteolytic activity in the epithelialized tissue by means of the tetracycline compound, most commonly decreasing collagenolytic activity and/or decreasing gelatinolytic activity. Collagenase and gelatinase activity have been shown to be decreased by the method of the invention. Preferably, the method is employed to improve the wound healing capacity of human or animal subjects in whom such capacity is impaired. Also, the non-antibiotic tetracycline is preferably administered topically at the site of the wound.
Cette invention se rapporte à un procédé permettant d'accroître la réaction de cicatrisation de tissu épithélialisé (par exemple, la peau, les muqueuses) à la suite d'une lésion traumatique aiguë. Ledit procédé consiste à utiliser un composé à base de tétracycline, pratiquement dépourvu d'activité antibactérienne, afin d'accroître la capacité du tissu épithélialisé à cicatriser des lésions aiguës. Spécifiquement, ce procédé consiste à accroître la vitesse d'accumulation du collagène du tissu épithélialisé jusqu'à un niveau supérieur à celui correspondant à la cicatrisation chez le patient. Ce procédé réduit l'activité protéolytique dans le tissu épithélialisé au moyen du composé à base de tétracycline, le plus souvent en réduisant l'activité collagénolytique et/ou l'activité gélatinolytique. On a démontré que le procédé de cette invention permet de réduire l'activité de la collagénase et de la gélatinase. De préférence, ce procédé sert à améliorer la cicatrisation chez des sujets humains ou animaux présentant un affaiblissement de cette capacité. En outre, on administre de préférence la tétracycline non antibiotique, de manière topique, au niveau du site de la lésion.
