Hammerhead ribozyme derivatives termed circular, hairpin, circular/hairpin, lariat, and hairpin-lariat ribozymes are disclosed. These hammerhead ribozyme derivatives are catalytically active and have increased specific activity and reduced requirement for divalent metal ion co-factor as compared to wild type hammerhead ribozyme. These derivatives can be used as antisense therapeutic compositions.
L'invention concerne des dérivés du ribozyme à tête de marteau appelés ribozymes circulaires, ribozymes en épingle à cheveux, ribozymes circulaires/en épingle à cheveux, ribozymes en lasso, ribozymes en épingle à cheveux/lasso. Lesdits dérivés du ribozyme à tête de marteau ont une activité catalytique, ainsi qu'une activité spécifique accrue et une exigence réduite d'un cofacteur d'ions métalliques bivalents, par comparaison avec le ribozyme à tête de marteau de type sauvage. Ces dérivés peuvent être utilisés en tant que compositions thérapeutiques antisens.
